Figure 2 - In the adipocyte, leptin inhibits lipogenesis by negative regulation of acetyl-CoA carboxylase, the rate limiting step in fatty acid synthesis. Leptin stimulates hormone sensitive lipase, which hydrolyzes triglycerides, diacylglycerols and cholesterol esters and downregulates lipoprotein lipase, reducing fatty acids available for lipogenesis.  Glucose uptake is inhibited by leptin, reducing the availability of glycerol-3-phosphate required for triglyceride synthesis.
